Work to provide the 10,000 new homes, a new entertainment and sports arena and leisure facilities on the Greenwich Peninsula will start this year.
The regeneration of the south-east London area will create about 24,000 permanent new jobs over 20 years.
Work on the homes is to start in 2006 and the Dome's new arena should be completed by spring 2007.
The agreements have been signed between Meridian Delta Ltd (MDL), English Partnerships which owns the Dome site, the Anschutz Entertainment Group and the London Borough of Greenwich.
Development of the site should finish in 2007
MDL director Bert Martin said: "We are determined that the Greenwich Peninsula will set new benchmarks in urban mixed use development and become one of London's finest districts, providing a unique blend of entertainment, business, residential and community uses."
Dome operator AEG is the largest owner of sports teams in the world with its portfolio including the Los Angeles Kings ice hockey team and the Los Angeles Lakers basketball side.

Although the Dome attracted more than six million visitors, its finances were based on 12 million visits and organisers had to continually ask for more lottery funds from the Millennium Commission. |
